Melanotan I

Target: Melanocortin 1 receptor (MC1R)

Melanotan I (Afamelanotide) is a synthetic analog of alpha-MSH that stimulates melanin production for skin tanning and photoprotection. FDA-approved for erythropoietic protoporphyria (EPP), a rare genetic condition causing extreme sun sensitivity. More selective than Melanotan II with a cleaner side effect profile.

Melanotan II

Target: Melanocortin 3, 4, 5 (non-selective)

Melanotan II is a synthetic peptide that stimulates melanin production (tanning) and has effects on sexual function and appetite. More potent than Melanotan I with additional effects on libido. Not FDA-approved; used in research settings.

PT-141

Target: Melanocortin 3/4 receptors (MC3R, MC4R)

PT-141 (Bremelanotide) is a melanocortin receptor agonist FDA-approved for hypoactive sexual desire disorder (HSDD) in women. Unlike PDE5 inhibitors, it works centrally in the brain to increase sexual desire rather than just blood flow. Originally derived from Melanotan II, it activates MC3R and MC4R receptors involved in sexual arousal and behavior.
