- PT-141 and PDE5 inhibitors (sildenafil, tadalafil, vardenafil) target entirely separate systems. PDE5 inhibitors block phosphodiesterase type 5, an enzyme that degrades cyclic GMP in smooth muscle cells of the corpus cavernosum.
