A 2020 comparative study in Journal of Immunology measured serum IL-6 levels in mice treated with equimolar doses of thymosin alpha-1 versus CpG ODN—CpG produced IL-6 elevations 4.8 times higher, yet both compounds achieved comparable dendritic cell maturation as measured by CD86 expression. This suggests thymosin alpha-1 selectively activates maturation pathways without triggering the full inflammatory cascade associated with stronger TLR-9 agonists.
