What if my lab's assay results don't match published thymosin alpha-1 data — did I do something wrong?

Verify peptide sequence and purity first, then reconstitution protocol. The most common cause of non-reproducible results with thymosin alpha-1 is receiving a truncated or substituted sequence that retains immunogenicity (triggers antibody response) but lacks TLR-2 binding capability. The mechanism through which it enhances dendritic cell IL-12 production and CD8+ T-cell activation. Request MS/MS sequencing from the supplier if not already provided. What if the peptide I received looks cloudy after reconstitution — is it contaminated?

Discard it immediately and contact the supplier. Cloudiness in reconstituted thymosin alpha-1 indicates aggregation or precipitation. Either the lyophilized peptide was improperly stored (temperature excursion during shipping), the reconstitution buffer pH is incorrect (should be 6.5–7.5), or the peptide contains synthesis byproducts that precipitate in aqueous solution. Aggregated peptides cannot be rescued by re-lyophilization or filtration. The tertiary structure is irreversibly altered. Properly synthesized thymosin alpha-1 reconstituted in sterile bacteriostatic water or PBS should be completely clear and colorless.
